Metabarcoding is a high-throughput DNA sequencing technique that uses targeted genetic markers to identify multiple species simultaneously from complex environmental samples like water, soil, or air. By analyzing environmental DNA (eDNA) shed continuously by organisms—cells from skin, scales, mucus, or feces—it reveals community structure and biodiversity across depth and habitat without requiring direct observation.
What this means in real life
A scientist collects water from a river, extracts all DNA present, and sequences it in bulk. Instead of identifying each organism individually, the method instantly reveals which fish, insects, plants, and microbes live there by matching DNA fragments to a reference database.
What it isn’t
It is not the same as identifying a single organism's full genome. Metabarcoding uses only tiny, standardized DNA snippets to quickly survey many species at once, rather than sequencing an entire organism's complete genetic code.
Commonly misused online
Often conflated with 'DNA barcoding' (which identifies one species at a time). Social media posts sometimes claim metabarcoding can identify every organism perfectly, ignoring that accuracy depends on reference databases and contamination.
